HoneyPal  CAP

Rapid Test for the presence of Chloramphenicol in honey

PRINCIPLE

The HoneyPal CAP is competitive test involving one receptor in one single operation, the acetamiprid in the sample compete for the antibody with the antigen coated on the membrane of the test cassette. Then after a color reaction, when the sample is free of chloramphenicol, a colour development occurs at the specific capture lines, indicating the absence of the concerned analytes in the sample. On the contrary, the presence of chloramphenicol in the sample will not cause the coloured signal to appear at the specific capture lines.

APPLICATIONS

The HoneyPal CAP is a rapid test that detect simultaneously the presence of chloramphenicol molecules in a honey sample.

Honey: 0.1 μg/kg (ppb)


SAMPLE PREPARATION
 Weigh 2.0±0.05g honey sample into a 15ml polystyrene centrifuge tube,add 2ml of Sample Diluent(Kit provided), shake with shaker to dissolve the honey thoroughly;
 Add 4ml of Sample Extraction Solution(Kit provided), shake intensively up and down for 5min,
 Centrifuge for Separation: 5 min / 3000g / room temperature (20-25℃);
 Transfer 3ml of upper layer of organic phase to a 10ml dry glass tube, dry with 50-60℃ water bath under nitrogen gas flow. Then dissolve the dry leftover with 0.2ml of Sample Buffer(kit provided), vortex for 1min, the solution is ready for use.

REMARKS: Proceed the assay within 60min.



TEST PROCEDURE

1. Read the instruction attentively, choose a clean and dry place to perform the test, wash and dry your hands before starting.
2. Take the kit out of the fridge and wait until the temperature of the reagents reaches the ambient temperature.
3. Calculate how many honey sample will be tested and mark the sample tubes to identify.
4. Take out as many test cassettes as the honey samples to be tested, and make proper marks to match the honey sample.
5. Pipette 75μl of the prepared sample solution into the orifice.
6. Start the timer to incubate for 10 minutes at room temperature (25℃);
During the incubation, The receptors will detect the presence of the antibiotics in the honey sample. The reaction will take 10 minutes.
7. Interpret the result.
File the used test cassettes if necessary.



RESULTS INTERPRETATION

Visual Interpretation

The cassettes are embodied with 2 lines, Control line, Test Line, and which are briefly expressed as “C” and “T”. The test results will be determined by the color presence of the lines. The following diagram describes the result identification.

NEGATIVE: C line, T Line are all red, which means that, the honey sample contains fewer antibiotics than value given in the table.
POSITIVE: C Line is red, T Line is invisible, which means that the honey sample contains the chlramphenicol more than the value in the table.

REMARKS: Control line is used as a quality indicator, which will always appear regardless of the T line. If Control line does NOT appear, it indicates that the result is invalid. Users please check the kit insert again and re-do the assay with new test cassettes.






STORAGE

2-8℃ in cool dark place, do not freeze. The kit will be valid in 12 months. The lot number and expired date are printed on the package.



PRECAUTIONS

 Do the assay following the instruction, do NOT touch the membrane of the cassette.
 Do NOT used the solutions provided from different batch in one assay.
 The cassette is used for only once; Do NOT use it repeatedly.
 The positive result should be further confirmed.
